PCC 6803 cells were grown at 30 °C in BG-11 media (Allen 1968) buffered with 35 mM HEPES at pH 7.8. Five hundred-milliliter volumes in 1-L flasks were bubbled with sterile-filtered air under continu- ous illumination of approximately 50 μEm−2 s−1 provided by fluorescent bulbs and grown to late log phase prior to harvesting and encapsulation.
