The modified BG11 was also buffered with 20 mM TES-KOH at pH 7.0, to inhibit the formation of HCO3-. The pre-cultures of the wild-type and mutant cells grown at logarithmic phase under CO2-enriched conditions (3% in air, v/v) were used to inoculate the experimental cultures at starting cell densities with an OD750 nm of 0.2. A relatively mild Ci stress was applied by switching the aeration from 3% CO2 in air (v/v) to air alone (350 ppm CO2, Ci-limited). The wild-type and mutant cells of Synechocystis 6803 were grown photoautotrophically at 32 °C (water bath) in BG11 medium buffered with 20 mM TES-KOH at pH 8.0, and bubbled with 3% CO2 in air (v/v).
